In vivo magnetic resonance diagnostic surrogate markers in prion diseases

Objective

The aim of the proposal is to identify in viva diagnostic and disease progression markers of transmissible spongiform encephalopathy (TSE) using non invasive brain magnetic resonance spectroscopy (MRS) and imaging (MRI) . Patients with suspected sCJD/vCJD or inherited forms of TSE, and patients with not-prion related dementiasand two different animal models of prison disease will be investigated using a multimode MR methodology. Main feature of the proposed MR method consists in the collection of quantitative MRI and MRS data and implementation of statistical analysis methods that will provide a highly sensitive technique for determining regional differences in tissue characteristics. The study of the animal models will identify MRI/MRS and pathological correlates in infected animals in order to provide fundamental information for the understanding of the pathological basis of brain MRI/MRS changes detected in TSE patients.
