Public website: the website will present the partners, the overall research goals and objectives and regular updates about achievements (publications, patents), ongoing events, symposium and related activities from other entities around the world that are of interest.
VIROFIGHT communication kitVIROFIGHT communication kit: This deliverable covers project flyer, slide show presentation (powerpoint, keynote) and poster.
Report on polymerized nucleic acids as engulfing shells on virusesTriangular subunits for prefabricated shells furnished with virusspecific aptamers such that shell assembly will be templated by a virus surface AU will design small nucleic acid tiles featuring multiple aptamers that bind a repetitive subunit in the viral surface to yield 24 multivalent 46 nm shell building blocks with sticky DNA linkers stabilized into duplexes by the avidity effect from the viral surface CryoEM and electrophoretic mobility shift assays will be used to assess particle shape integrity and proper basic functionality Designs will be iteratively improved Report will cover results of these actions
Generation of non-infectious and infectious virus particles from non-enveloped virusesGeneration of noninfectious and infectious virus particles from nonenveloped virusesThis deliverable comprises solutions containing virus particles to be handed over to partners of the consortium for further experiments
Report on library of prototype DNA origami shell designsReport on library of prototype DNA origami shell designs: This report will contain blueprints, 3D models, and DNA sequences needed to build the shells design variants. In addition, wherever applicable, experimental data obtained on the designs will be added and discussed, comprising e.g. cryo EM images, electrophoretic mobility shift analysis. etc
Generation of VLPs and LVs pseudotyped with surface proteins of enveloped virusesGeneration of VLPs and LVs pseudotyped with surface proteins of enveloped viruses: This deliverable comprises solutions containing the virus-like particles and lentiviral pseudotyped particles to be handed over to partners of the consortium for further experiments.
Report on assembly procedures for prefabricated DNA origami shellsReport on assembly procedures for prefabricated DNA origami shellsIn this report experimental procedures will be described leading to successful assembly of prefabricated DNA origami shells including thermal annealing conditions reaction mixture conditions purification steps and quality control procedures In addition troubleshooting measures and inluence of environmental conditions such as temperature or cation content on assembly will be described
Dissemination and communication planDissemination and communication planA detailed communication and dissemination plan covering all communication and dissemination activities required during the project and adapted to the different target audiences and objectives of the communication or dissemination measures
Generation of hepatitis B viruses as exemplary enveloped virusGeneration of hepatitis B viruses as exemplary enveloped virusThis deliverable comprises solutions containing the hepatitis B viruses to be handed over to partners of the consortium for further experiments
Exploitation Innovation PlanAn initial exploitation innovation plan will be presented in M12 An updated version will be created in M36
Design details and protocols for modular swap out of virus recognizing moietiesDesign details and protocols for modular swap out of virus recognizing moietiesSubsets of shellforming DNA oligonucleotides will be modified with terminal extensions pointing into the shell interior The sequences of the extensions will first be dummies to deal with and remove potential aggregation issues induced through Velcro effects Iterative refinements will be made so that arbitrary sequences can be attached in tunable decoration density and geometry in a plug and play fashion without affecting the shell quality and functionality AU will deliver virusbinding candidate DNA aptamer sequences to be included as direct terminal extensions or sticky linkers
