Individuation of indicators at different biological levels for early prediction of the onset and fate of skeletal anomalies in model species for marine (medaka) and freshwater (zebrafish) commercial fishESR4 Evaluation of skeletal anomalies requires a welldefined classification of the observed defects and an objective identification of the various types of defects
Primary effect of inappropriate P supply and function of Fgf23 on growth and development of bone tissue in Atlantic salmon.ESR12 Phosphate P is a limiting resource directly related with organic pollution whereas unbalanced diets may affect animal health and welfare with occurrence of anomalies P content and efficiency in fish feeds based on plant ingredients is frequently low A phosphorus deficient vertebral phenotype in juvenile salmon will be used to test the effect of different factors on bone in an in vivo model
Technical articlesOverview articles presenting and analysing combined results of several ESRs
Evaluation of risk and prediction of skeleton anomalies.ESR7 To precisely assess the burden of skeleton anomalies in European fish farms and to determine the socioeconomic impact regarding the prevention and treatment of skeleton anomalies in fish farms two main methodologies discrete choice analysis and risk assessment will be used
Biomedaqu Final workshop. All exploitation plans completed.Final workshop and final reports All activities finished
Physiological mechanisms of Zn, Mn and Se effect on bone development and mineralization.ESR6 Sustainable development of aquaculture asks for high quality ingredients to replace fishmeal and fish oil and fulfilling nutritional requirements This replacement presently leads to several skeletal alterations suggesting nutritional imbalances eg Zn Mn and Se are lower in plant ingredients Optimal dietary levels of Zn Mn and Se for skeletal health in seabream will be determined d14 M40 and the physiological mechanisms investigated through mutant fish models

Satellite section in European Aquaculture Society Conference.Organisation and participation of a satellite meeting at the Conference of the European Aquaculture Society
Predictions of the effect of preventive measures when applied by fish farmers.Discrete choice experiments DCE have been used in many research fields to elicit preferences of decision makers regarding different products and services They involve the creation of hypothetical scenarios trying to mimic real markets which are defined to answer different research questions Thus it is possible to analyze the effect of important attributes over the outcomes of the experiment to quantify tradeoffs between attributes and to evaluate the willingness to pay for improving the quality of a product or service In our case different skeleton anomalies in form and severity will be defined and used Data will be obtained from a large number of companies and consumers within the consortium and through contacts from FEAP and API DCE provide rich data sources which have been widely used in health economics to evaluate preferences for healthcare products and services they allow researchers to obtain after the application of appropriate modelling methodologies such us discrete choice models merging their financial cost and revenues and their organizational consequences risks and controls to implement It will also provide essential inputs for the main economic evaluation techniques costbenefit analysis CBA and costutility analysis notably when coupled with riskcontrol matrix ULGCEPE
Dissemination and Outreach plan including website.Disseminate project activities and results (d6.1, M4), both to professionals (fish farmers, stakeholders, researchers) and the public. Public reports from the different WPs will also be made available through the site. A page on a social network (i.e., Facebook) will be created and coordinated, directed for a larger non-scientific audience
Practical recommendations for fish farms and feed producers on vitamins K and D dietary levels to avoid bone anomalies and pathological effects of Ca metabolism alterations.ESR8 Fatsoluble vitamins K VK and D VD play essential roles in Ca transport and tissue deposition and hence in bone development mineralization and remodelling Excessive VK and VD dietary levels can lead to bone anomalies there is a large variation in the content of these vitamins in live preys fed to marine fish larvae and their optimum levels during larval stages have not been yet determined for marine fish
In vivo and in vitro systems to test osteogenic activitiesAt the molecular level, skeletal formation and homeostasis depends on the presence of specific cells and their differentiation, proliferation and function in skeletal tissues influences extracellular matrix (ECM) and hydroxyapatite deposition. Osteoblast- and chondroblast-like skeletal derived cells from teleosts will be used as experimental model systems to assess the effects of micronutrients, at the cellular level, on the capacity to mineralize the ECM
BioMedAqC Supervisory Guidelines and ETN Training program.Set up SB, Recruitment guidelines, Satellite Meetings and supervision infrastructure
Description of cellular basis of spine repairESR14 Scoliosis a deviation of the normal line of the spine is one the main ecological threats to commercial fish farming Hence understanding what leads to malformations of the axial skeleton in fish is not only of scientific interest but also has considerable economical relevance The complete understanding of the underlying biology has been hampered by an inability to clearly identify the cells which are responsible for mineralizing the axial skeleton and by an inability to follow the dynamics of axial skeletogenesis at the level of gene expression We have recently shown that the extracellular Entpd5 an enzyme providing phosphate for the mineralization process is expressed in a segmental manner within chordoblastsnotochord sheath cells of the notochord Lleras Forero submitted and have furthermore demonstrated that these cells have the propensity to repair damage inflicted to the notochord Lopez Baez submitted Based on these findings it will now be possible to understand in detail how expression of Entpd5 is orchestrated by the embryo to form the larval axial skeleton during physiological conditions and to repair skeletal elements upon pathophysiological damage
2 oral communications /student (30).Publications At least two technical articles are planned for publication in professional newspapers eg Fish Farming International andor magazines eg World Aquaculture or Aquaculture Europe summarizing the main results and outcomes of the project other than the scientific publications described in WP1

Function of new candidate genes in zebrafish bone formation. Relevance for human (and general) skeletal pathology.ESR23 Zebrafish models have been proven relevant to study skeletal pathologies encountered in humans such as osteoporosis or scoliosis but possibly also in farmed fish ARS has identified the FIBULIN3 protein to be upregulated in serum of osteoarthritic patients UPD7 has identified several variants of genes in cohorts of young patients with osteoporosis osteoarthritis or ectopic mineral deposits in the cartilage such as chondrocalcinosis The function of these factors will be evaluated at the wholebody level by generating the zebrafish mutants in the genes of interest eg fbln1 fbln3 il11ra lrp5 wnt1 iftim5 plekhm1 using the CRISPR methodologies The mutant fish phenotype is characterized using established skeletal staining methods fluorescent transgenic lines microCT histomorphometric methods and by determining the expression of marker genes by in situ hybridization or RTPCR Variant proteins will be tested for proliferation in human and fish osteoblast cell culture Genes affecting skeletal development will be characterized in aquaculture species and their expression will be followed in challenging conditions that generate skeletal malformations including exposure to altered gravity conditions to evaluate their contribution to the defects

Supervisory BoardSupervisory board recruited.
Organisation and participation to the 6th IAFSB conference
ESR1: Based on previous works and open-source tools to identify skeletal features in the head of 9-days old normal zebrafish larvae, we will develop an interactive tool to automatically identify, annotate and quantify skeletal elements in different types of images from fish. The tool will be first calibrated for analysis of normal and deficient zebrafish larvae, before extending to other types of skeletal images (fluorescent, X-ray), also of other species. Tools will be implemented for measuring sizes, relative positions and density of bone elements. Finally, a reporting tool including statistical analysis will be developed. Dissemination of these tools within the consortium and among the wider community will be implemented with the help of CTM
An BioMedAqC web page will be prepared to disseminate project activities and results
