RNA-codified release of cytotoxic peptides from PNA prodrugs as a new therapeutic approach to cancer

Objective

In this proposal we intend to develop a new medicinal chemistry concept base on cell-personalized delivery of tailored drugs. Relying on basic chemical tools such as proximity-induced acyl transfer reactions and catalytic turnover- we will use RNA biomarkers of cancer cells to trigger the specific expression of cytotoxic drugs. This approach, in addition to contribute to basic science in chemistry and chemical and cell biology and suggest new therapeutic strategies, might allow to overcome the most important observed complications in cancer treatment: selectivity and resistance.
