Microbial marine communities diversity : from culture to function

Objective

This proposal investigates marine microbial biodiversity by obtaining cultures of novel microbial groups of marine origin, measure their functional origin and using molecular identity indicators. We plan a systematic approach that includes scale screening of culture conditions using microtiter plates equivalent to 100000 classical experiments of liquid media enrichments, coupled to extinction dilution to guarantee that at least all the main physio-ecotypes have a chance to grow. The vast database of the organisms that develop under combinations of nutrient regimes will be of critical value in making rapid progress to describe marine microbial biodiversity. Many non-conventional techniques to obtain pure cultures in a single step will be applied by the use of genetic manipulation or flow cytometry. A collection of several thousand strains will be obtained which will be analysed for their role in the ecosystem functioning.
