Deliverables

Report on the relationships of early-life exposures with trajectories of cardiovascular and metabolic risk factors from birth to adulthood

We will study the associations of individual earlylife stressors and the integrated earlylife exposome with cardiometabolic phenotypes at various ages across the life course and with trajectories of those phenotypes

Report on differences in DNA methylation loci that mediate the associations of early-life exposures with mental health and psychopathology life course trajectories

We will identify DNA methylation loci that mediate the relationships of earlylife stressors and the integrated earlylife exposome with mental health and disease trajectories

Report on the associations of DNA methylation loci related to early-life stressors with functional outcomes, including RNA expression

We will explore associations of differences in DNA methylation at specific CpG sites discovered in EWAS with RNA expression levels methylationtranscription analyses using RNA expression data measured in some of the participating cohorts at various ages in childhood

Report on impact of early-life policies and interventions based on dynamic microsimulation models

Dynamic microsimulation modelling involves the generation of simulated data on individual units and is particularly suited for policy evaluations and scenario analyses It is a multiprocess simulation model in which each process is estimated at an individual level and feeds back into the other processes In LIFECYCLE the analytical framework and the specific models will be informed with populationspecific exposure prevalences and effect estimates of distal earlylife stressors and intermediates obtained from the EU Child Cohort Network and external data

Report on the associations of early-life stressors during preconception, pregnancy, infancy and early childhood with lung function developmental trajectories and the risk of asthma from childhood until young adulthood

We will use individual participant data from the EU Child Cohort Network to identify earlylife stressors individually and integrated in the earlylife exposome with lung function trajectories and symptoms diagnosis and medication related to asthma Main outcomes include lung function FEV1 FEV1FVC FEF2575 FEF75 bronchial hyperresponsiveness and symptoms and diseases respiratory infections wheezing asthma

Report on internalizing and externalizing behavior and other mental health trajectories from birth to childhood, and their associations with psychopathology outcomes

We will model longitudinal life course trajectories of mental function from early life to adulthood using data from the EU Child Cohort Network These trajectories will include both tracking and stability of internalizing and externalizing behavioural indicators measured with the Child Behaviour Check List CBCL throughout childhood and the relationships of these behavioural indicators with psychopathology indicators in adolescence and adulthood

Report on associations of early-life stressors with DNA methylation at birth

We will use an EWAS approach to study the associations of the main indicators of early-life stressors and the integrated early-life exposome, before and during pregnancy on DNA methylation at birth measured in cord blood. We will also explore the associations of the overall early-life exposome, developed in WP3, during pregnancy, on cord blood methylation at birth.

Report on review of interventions focused on lifestyle stressors, to improve life course health trajectories

We will perform literature searches and reviews based on the developed strategies from deliverable 9.1. We will review evidence focused on interventions on lifestyle stressors of parents and young children. In line with WP3, these stressors will include parental smoking, alcohol consumption, illicit drug use, sleep, physical activity and parent and childhood nutritional intake and status.

Report on exposome models based on proof-of-principle studies

We will develop protocols for analysis of associations of a fully integrated earlylife stressor model the earlylife exposome with lifecycle health outcomes and test these in proofofprinciple studies As input we will use the main earlylife stressors WP1 WP3 Deliverable 31 34 Proofofprinciple datasets will be those of the EUFP7 funded HELIX project wwwprojecthelixeu including six cohorts across Europe n1200 plus the Generation R Study n7000 and DNBC n70000

Report on review of interventions focused on migration stressors, to improve life course health trajectories

We will perform literature searches and reviews based on the developed strategies from deliverable 9.1. We will review evidence on interventions on stressors related to migration. In line with WP3, the stressors will include indicators at individual, neighbourhood and macro-levels such as country of origin of the individual, its parents, or grandparents, generation after migration, measurements of integration.

Report on approaches to analyse multiple stressor data in the context of longitudinal modelling

In WP3 we will develop models to analyse associations of multiple stressors many of which are multidimensional with health outcomes This task will further develop these methods by assessing approaches to analyse multiple correlated stressor data in the context of longitudinal modelling

Report on the persistence of DNA methylation differences related to early-life stressors

To establish the role of DNA methylation differences on later life health outcomes in more detail and to better understand how these differences may exert effects, we will examine DNA methylation differences from birth to early adulthood in relation to early-life stressors.

Report on review of interventions focused on urban environment stressors, to improve life course health trajectories

We will perform literature searches and reviews based on the developed strategies from deliverable 9.1. We will review evidence focused on interventions on urban environment stressors. In line with WP3, these stressors will include measurements of air pollution, noise, green space, connectivity and walkability.

Report on the effects of early-life stressors on mental health and psychopathology life course trajectories

We will use data from the EU Child Cohort Network to study the associations of earlylife stressors including socioeconomic migrant urban environment and lifestyle related indicators and the integrated earlylife exposome with mental development mental health and risk factors for psychiatric diseases at various ages across the life course

Recommendations for public health strategies focused on future parents and young children to improve life course health trajectories from early life onwards

We will develop recommendations for public health strategies based on findings from LIFECYCLE and systematic reviews of literature on interventions about modifications of earlylife stressors WP9

Report on early-life stressors that are related to both asthma throughout childhood and lung function and COPD in adulthood

We will explore whether individual earlylife stressors and the integrated earlylife exposome related to impaired respiratory health at younger age also affect lung function and risk of COPD in adulthood

Report on review of interventions focused on socio-economic stressors, to improve life course health trajectories

We will perform literature searches and reviews based on the developed strategies from deliverable 9.1. We will review evidence focused on interventions on socio-economic inequalities In line with WP3, socio-economic related stressors will include parental education, occupation, type of employment contract, family income, family size, child care and house property and size.

Report on cardiovascular developmental trajectories during fetal life, infancy and early childhood, and the relationship with early-life exposures and cardiovascular risk factors in childhood

We will use data from the EU Child Cohort Network partners that collected data before or during pregnancy and have detailed fetal and infant growth and cardiovascular development data. We will study associations of early-life stressors and the early-life exposome with adiposity and cardio-metabolic developmental trajectories in fetal life, infancy and early childhood (fetal and placental blood flow, cardiac and vascular dimensions, adiposity), and subsequently associations of these trajectories with risk factors for childhood for cardio-metabolic disease.

Report on the relationship of early-life exposures with differences in metabolic profiles and the extent to which these mediate associations of early-life exposures with cardio-metabolic risk factors and disease

We will explore the associations of individual earlylife stressors and the integrated earlylife exposome with a wide range of metabolites reflecting systemic metabolism that might mediate associations of earlylife exposures with subsequent cardiometabolic disease

Report on various life course models for the effects of early-life exposures on later cardio-metabolic risk factors and disease

In line with deliverable 41 and WP7 we will compare various potential life course models to best explain the relationships of earlylife stressors and potential mediators with trajectories of cardiometabolic risk factors and disease All EU Child Cohort Network cohorts will contribute to one or more of these trajectories

Report on DNA methylation changes as measures of early-life exposures

We will explore if DNA methylation patterns can be used as markers for earlylife stressors Building on deliverables 8184 we will assess if DNA methylation sites or patterns that are related to earlylife stressors are able to accurately identify those exposed ie specific enough to the exposure in question

Report on to what extent differences in DNA methylation mediate the associations between early-life stressors and subsequent life course chronic obstructive respiratory health and disease trajectories

We will identify DNA methylation loci that mediate the relationships of earlylife stressors with chronic obstructive respiratory health and disease throughout the life course We will use data from studies that have epigenomewide data on DNA methylation which can be used for epigenomewide association studies EWAS metaanalyses to identify epigenetic markers associated with respiratory outcomes

Integrated summary report on recommendations on potential interventions focused on modification of early-life stressors

We will integrate results from the reviews from deliverables 92 95 for recommendations for feasible effective and efficient interventions before or during pregnancy in infancy or in early childhood Specific subgroups of interest include socioeconomic and ethnic groups or subgroups identified at increased risk based on other parental or infant characteristics

Report on the relationships of early-life stressors with transient and persistent common allergies, and the extent to which these mediate associations of early-life stressors with life course chronic obstructive respiratory health and disease trajectories

We will relate individual earlylife stressors and the integrated earlylife exposome with common allergies from birth to young adulthood We will also examine if we can identify subgroups based on allergic and nonallergic pathways

Report on differences in DNA methylation loci that mediate the associations of early-life exposures with cardio-metabolic risk factors and disease

We will identify DNA methylation loci that mediate the associations of individual earlylife stressors and the integrated earlylife exposome with cardiometabolic risk factor trajectories and disease We will use data from studies that have epigenomewide data on DNA methylation which can be used for epigenomewide association studies EWAS metaanalyses to identify epigenetic markers associated with cardiometabolic outcomes

Report on approaches for longitudinal data/health trajectories in the context of life course analyses

We will provide a detailed description of analytical options to analyse repeated measurements data. We will cover theoretical biostatical aspects, the link between statistical modelling and the underlying biological frameworks and more practical applications dealing with issues of implementation and interpretation.

Report on patterns of core and integrated socio-economic, migration, urban environment and lifestyle related stressors across Europe

We will use all core and integrated data from WP3 deliverables 3.1 - 3.4 and apply factor analysis and model-based latent class analysis to uncover the nature of the underlying dependencies between stressors and to define stressor patterns. Participating partner cohorts are those that have collected key variables in each of the areas. Particular attention will be paid to comparing of stressor patterns between regions

Report on approaches for causal inference in the context of life course trajectory analyses

We will develop an analysis plan providing a detailed description of options in study design and statistical analyses to apply causal inference methods to life course research. This will integrate theoretical and applied aspects, focusing on implementation and interpretation.

Report on the impact of early-life stressors on population and individual economic outcomes

We will identify links between earlylife stressors and economically related later life outcomes and elucidate mechanisms underlying these links We will use longitudinal data from the EU Child Cohort Network by linking earlylife stressors to economicallyrelated outcomes in childhood adolescence and adulthood Next we will use registrybased data from Statistics Netherlands CBS and Statistics Finland to relate earlylife stressors such as recessions to economically related outcomes later in life

Report on the relationship of early-life stressors with fetal and childhood brain developmental outcomes and the extent to which these mediate associations of early-life exposures with mental health and psychopathology life course trajectories

We will study brain development from the fetal period and childhood onwards by means of advanced imaging studies including repeated fetal ultrasound and childhood MRI brain studies We will first study the role of brain developmental adaptations in fetal life or childhood Generation R Generation R Next INMA ALSPAC BIB NFBC1986 RAINE and subsequently try to replicate the findings in adults NFBC1966 This deliverable will provide insights to the role of developmental structural and functional brain developmental adaptations in the relationships between earlylife stressors and mental health trajectories

Personalized prediction models based on early-life stressors to predict development of cardio-metabolic, respiratory and psychiatric disease

We will use data from the EU Child Cohort Network to develop models (1) to predict from pregnancy and early childhood stressor data the onset of risk factors for cardio-metabolic, respiratory and mental outcomes throughout the life course (infant prediction models) and (2) to predict persistence of childhood cardio-metabolic, respiratory and mental outcomes (childhood prediction models).

Training course on causal inference methods and longitudinal modeling in the context of life course trajectory analyses

The courses will be developed to answer the main demands for theoretical and applied knowledge emerging from the interaction with WP4, WP5 and WP6.

E-learning module available online

We will develop an e-learning module, consolidating the scientific evidence base and status of recommendations of LIFECYCLE topics and outcomes. This e-learning module will be targeted to international AHPs and researchers working in the field and will be implemented and made publically available on the ENeA platform without access restriction (www.enea.moodle.elearning.lmu.de).

Tutorials on the implementation of causal inference methods and longitudinal modelling in the context of life course trajectory analyses

We will develop, in collaboration with WP4, WP5 and WP6, tutorials on selected outcomes to demonstrate the implementation of the analysis strategy in specific contexts. The main focus of the tutorials will be on the comparison of approaches and the interpretation of the resulting estimates.

eHealth applications for personalized prevention strategies

We will maximize the impact of the main results for individuals by developing a website and mobile applications for individually customized advice to women and men before pregnancy and for pregnant women and young children

External project website launched

Project website accessible for general public and specific public. The website will be multi-layered. The homepage will be accessible to everyone; it will be based mainly on visual communication (videoclips, data visualisations, etc.); and it will link to specific pages for various stakeholder or target groups, including researchers, health care providers, patient organizations, patients and policymakers.
