The overall objectives of the INTEGRA project is to develop new genetic engineering tools that will permit targeted integration of a transgene into the genome of the desired eukaryotic organism. This project will allow the easy and rapid generation of transgenic knock out or knock in small and large animals without the need of embryonic stem cells. This will have visible repercussions on fundamental and applied research particularly in pharmaceutics agronomy and food sciences.
At the same time it will strengthen European research in the field of gene therapy with the development of safe viral vectors based on non integrative or targeted integration vectors adequate for clinical use. We plan to meet the objectives by two alternative means. The first one will combine the use of a lentiviral episomal vector with a phage site specific integration system to direct the integration of the transgene into a unique locus in the host genome. The alternative method will use lentiviral vectors for which the integrase specificity will be modified HIV integrases will be generated whose DNA binding domain will be replaced by a synthetic DNA binding domain able to recognize any given DNA sequence of 9 or 18 base pairs. Such specific integrase will be incorporated into a gene transfer vector system that will allow the genetic modification of somatic cells and zygotes.
These technologies will be used by the partners to generate transgenic livestock with modified traits improved growth traits disease resistance and to generate sensor animals to monitor contamination of livestock by toxic agents. On the other hand gene therapy preclinical studies will be performed using the secured vectors Specific ethical and societal implications of the project will be anticipated through the participation of European stakeholders. The overall INTEGRA project clearly cuts across the Thematic Priority 1 and 5 and the Science and Society programme of FP6.
