DNA methylation is an important epigenetic mechanism necessary for normal gene expression and cellular function. The mechanisms regulating its correct distribution throughout the genome are incompletely understood. Using protein interaction screens, we identified a new interaction between the maintenance methyltransferase Dnmt1 and Drosha, an important RNase enzyme involved in small RNA biogenesis. Drosha is necessary for full Dnmt1-mediated DNA methylation. Our findings support the notion that RNA plays an important role in in controlling Dnmt1 methyltransferase activity and therefore correct patterns of DNA methylation.
