Cytogenetic biomarkers and human cancer risk

Cel

The partners have previously shown that a high frequency of chromosomal aberrations (CAs) in peripheral lymphocytes is predictive of increased cancer risk, regardless of smoking or occupational carcinogen exposure. The present multidisciplinary project aims at clarifying the reasons for these findings. Archived cell specimens of subjects earlier examined for cytogenesis biomarkers will be analysed for major genetic polymorphisms suspected to affect chromosome damage. These data will be used to reveal those Polymorphisms that modify the frequency of CAs and of sister chromatic exchanges (SCEs) and the cancer association of CAs. The existing cohorts will be re-examined to identify the types of CAs indicative of cancer risk and to re-evaluate if SCEs predict cancer. Two new European cohorts will be assembled, one to confirm the findings on CAs and SCEs and to study association with major cancer types, and another to see if cancer risk is predicted by lymphocyte micronucleus frequency.
