The role of intercellular communication and DNA double strand breaks in the induction of bystander effects

Cel

The biological processes mediating bystander responses are incompletely understood despite the relevance of this phenomenon to radiation protection and radiation therapy. The present proposal contributes to the elucidation of the mechanisms underlying bystander effects by evaluating intercellular communication as a determinant of the response, and DNA double strand breaks as initiating lesions. Using biological model systems with conditional induction of intercellular communication and diverse experimental approaches, some of which are developed and tested here, we aim to investigate the induction kinetics of by stander effects in cultured cells and evaluate their contribution to the response of tumors growing in nude mice to ionizing radiation. The results should allow the definition of targets for pharmacological intervention to protect humans from the adverse effects of ionizing radiation.
